AGS-16M18 is a fully human monoclonal antibody developed as an investigational therapy for kidney and liver cancers. It targets the cell surface protein Ectonucleotide pyrophosphatase/phosphodiesterase family member 3 (ENPP3, also known as AGS-16), which is highly expressed in over 95% of kidney cancers, including clear cell and papillary carcinomas, and approximately 40% of hepatocellular carcinomas. The antibody binds to ENPP3 on tumor cells, inhibiting their proliferation, migration, invasion, and angiogenesis. Preclinical studies demonstrated that AGS-16M18 has potent antitumor activity both in vitro and in vivo by impacting key signaling pathways involved in proliferation (ERK, AKT) and apoptosis (PARP). The drug was developed using XenoMouse technology to generate a fully human IgG1λ monoclonal antibody with high affinity for its target. Clinical development focused primarily on advanced renal cell carcinoma; however, the program was discontinued after Phase 1 trials[1][2][3][4].
