AGS-1C4D4 is a fully human IgG1κ monoclonal antibody that targets the prostate stem cell antigen (PSCA), a cell-surface protein highly expressed in most prostate cancers and also present in pancreatic and bladder cancers. Developed originally by Agensys, this antibody was designed to exert antineoplastic effects primarily through antibody-dependent cellular cytotoxicity (ADCC) and immunostimulatory mechanisms. It was produced in Chinese hamster ovary cells and underwent clinical evaluation for metastatic castration-resistant prostate cancer as well as metastatic pancreatic cancer. Clinical trials demonstrated that AGS-1C4D4 was generally well-tolerated, with mostly mild adverse events, but development for both indications has been discontinued[3][6][7].
