AGSAV301

AGSAV301 is a fixed-dose combination (FDC) therapy developed by Ahn-Gook Pharmaceuticals for the treatment of hypertension. It consists of S-amlodipine, the active (S)-enantiomer of the calcium channel blocker amlodipine, and valsartan, an angiotensin II receptor blocker (ARB). S-amlodipine inhibits the influx of calcium ions into vascular smooth muscle and cardiac muscle, leading to vasodilation and reduced peripheral resistance. Valsartan selectively blocks the binding of angiotensin II to the AT1 receptor, inhibiting its vasoconstrictive and aldosterone-secreting effects. By combining these two mechanisms, AGSAV301 provides synergistic blood pressure lowering effects. It was developed as a potentially more potent or better-tolerated alternative to the racemic amlodipine/valsartan combination (Exforge), as S-amlodipine is the pharmacologically active isomer responsible for the antihypertensive effect of amlodipine while potentially reducing side effects like peripheral edema.
