AGT-010 is a selective, non-hydroxamic, non-oxadiazole small molecule inhibitor of histone deacetylase 6 (HDAC6) currently in IND-enabling development by Augustine Therapeutics. It is being investigated for the treatment of heart failure with preserved ejection fraction (HFpEF) and associated cardiometabolic dysfunction. Unlike pan-HDAC inhibitors, AGT-010 specifically targets the cytoplasmic HDAC6 isoform, which regulates α-tubulin acetylation and cytoskeletal dynamics. By increasing α-tubulin acetylation, AGT-010 improves cardiomyocyte compliance and diastolic mechanics. Preclinical studies in HFpEF models have demonstrated that AGT-010 treatment reduces left ventricular pressures, attenuates cardiac hypertrophy, and improves metabolic parameters including glucose tolerance and insulin sensitivity.
