Overview
AGX-148 is an enhanced tumor-infiltrating lymphocyte (TIL) therapy developed by AgonOx and SQZ Biotech for the treatment of malignant solid tumors, including melanoma. This adoptive cell therapy involves isolating and expanding highly enriched, tumor-reactive CD8 T cells from a patient's own tumor tissue—specifically those co-expressing CD103 and CD39 markers—to create a potent therapeutic product. These double-positive (DP) TILs demonstrate increased ability to recognize and kill autologous tumor cells both in vitro and in vivo. The mechanism of action includes immunologic cytotoxicity as well as functional replacement or enhancement of interleukin 12 (IL-12), interleukin 2 (IL-2), and T lymphocytes. Clinical development is ongoing, with phase I trials underway for various solid tumors[1][2][3][4][5][7].
