AGX101 is a first-in-class, nuclear-delivered antibody-drug conjugate (ND-ADC) developed to treat solid tumors by targeting TM4SF1 (Transmembrane-4 L-Six-Family-Member-1), a protein highly expressed in both tumor cells and tumor vasculature but with limited expression in normal adult tissues[1][2][6]. AGX101 consists of an antibody directed against TM4SF1, conjugated to a potent maytansinoid tubulin inhibitor payload. The unique internalization pathway of TM4SF1 enables the delivery of the cytotoxic payload directly into the nucleus of cancer and endothelial cells within the tumor microenvironment[1][2]. AGX101 exerts its anti-tumor effects through three mechanisms of action: direct killing of tumor cells, elimination of tumor blood vessels (depriving tumors of their blood supply), and activation of immune surveillance within the tumor microenvironment[1][8]. Preclinical studies have demonstrated a broad therapeutic window with no adverse clinical signs at efficacious doses, supporting its potential for treating various solid cancers. The drug is currently being evaluated in Phase 1 clinical trials for advanced solid tumors[4][5].
