AGX1053 is a first-in-class antibody-drug conjugate (ADC) developed by Angiex, designed to target Transmembrane 4 L6 Family Member 1 (TM4SF1). TM4SF1 is a protein highly expressed on the surface of various solid tumor cells and, uniquely, on the tumor-associated vascular endothelium, while remaining largely absent from normal resting vasculature. The drug consists of a humanized monoclonal antibody conjugated to the potent microtubule-inhibiting maytansinoid payload, DM4 (ravtansine), via a hindered disulfide linker. AGX1053 employs a dual-action mechanism: it directly kills cancer cells and selectively destroys the tumor's blood supply by targeting the activated endothelium of tumor vessels. This vascular-targeted approach is intended to enhance drug delivery and efficacy in poorly perfused solid tumors. It is currently being evaluated in Phase 1 clinical trials for patients with advanced solid tumors.
