Overview

AHT956 is a small molecule inhibitor of protein kinase C (PKC), developed by Novartis. It acts as a PKC inhibitor and has been investigated primarily for its potential in immune system diseases and skin/musculoskeletal diseases, with particular preclinical focus on uveal melanoma harboring GNAQ or GNA11 mutations. In preclinical studies, AHT956 demonstrated selective growth inhibition in melanoma cell lines with these mutations, inducing G1 cell cycle arrest and suppressing DNA synthesis. The drug reached Phase 1 clinical trials to assess safety, tolerability, pharmacokinetics, and pharmacodynamics in healthy subjects but has not advanced beyond early-stage development[1][2][4].
