Overview

AI-10-104 is a small molecule inhibitor that targets the transcriptional activity of the RUNX (Runt-related transcription factor) family. It functions by disrupting the protein-protein interaction between RUNX proteins (such as RUNX1 and RUNX2) and their essential co-factor, CBFβ (Core Binding Factor subunit beta). In preclinical studies of triple-negative breast cancer (TNBC), AI-10-104 has demonstrated the ability to reduce cell viability and migration, induce apoptosis, and hinder the formation of mammospheres, which are markers of cancer stem cell activity. Furthermore, AI-10-104 has been shown to enhance the toxicity of chemotherapeutic agents like doxorubicin and restore sensitivity to paclitaxel in resistant TNBC cells. It also appears to modulate the tumor immune environment by downregulating PD-L1 expression and affecting mitochondrial dynamics.
