Overview

Ai-TCR-T cells are an adoptive cell therapy consisting of T cells engineered to express an affinity-improved T-cell receptor (TCR) targeting the hepatitis B virus surface antigen (HBsAg). Specifically, these cells are designed to recognize the HLA-A*02:01-restricted HBs371-379 peptide. By enhancing the binding affinity of the TCR compared to wild-type versions, the therapy aims to overcome the low antigen expression and immune escape mechanisms common in HBV-related hepatocellular carcinoma (HCC). Preclinical studies have demonstrated that Ai-TCR-T cells exhibit improved sensitivity and cytotoxicity against HBV genotypic variants and can eliminate HCC tumors in xenograft mouse models without recurrence.
