Aib-CAR-T is an activation-inducible bispecific chimeric antigen receptor (CAR) T-cell therapy targeting B-cell maturation antigen (BCMA) and CD24 for the treatment of multiple myeloma. Developed to address the safety concerns of targeting CD24—which is expressed on various healthy tissues including neural, epithelial, and pancreatic cells—the Aib-CAR-T construct utilizes an "AND-gate" logic circuit. It consists of a constitutively expressed BCMA-CAR and an inducible CD24-CAR controlled by a nuclear factor of activated T cells (NFAT) promoter. Upon binding to BCMA on myeloma cells, the T cell is activated, triggering the expression of the CD24-CAR via the NFAT signaling pathway. This localized activation ensures that the CD24-targeted cytotoxic activity is confined to the myeloma microenvironment, effectively targeting CD24-positive stem-cell-like myeloma cells (critical factors for relapse) while sparing healthy CD24-expressing tissues.
