AIC100 is an affinity-tuned, third-generation chimeric antigen receptor (CAR) T-cell therapy that targets intercellular adhesion molecule-1 (ICAM-1) on tumor cells. It is designed to treat advanced thyroid cancers, including anaplastic thyroid cancer (ATC) and poorly differentiated thyroid cancer (PDTC), by redirecting autologous T cells to recognize and kill ICAM-1–expressing malignant cells. The CAR construct features a lower-affinity LFA-1 binder for ICAM-1, aiming to reduce toxicity while maintaining antitumor efficacy. Developed using AffyImmune’s Tune and Track CAR T cell platform, AIC100 has demonstrated promising safety and efficacy in early-phase clinical trials with manageable cytokine release syndrome and no observed neurotoxicity. The therapy has received FDA Fast Track, Orphan Drug, and Regenerative Medicine Advanced Therapy (RMAT) designations for the treatment of advanced thyroid cancers[4][5][6][7][8][9][10].
