Overview

AIC467 is a small molecule non-nucleoside polymerase inhibitor (NNPI) developed by AiCuris for the treatment of a broad range of herpesvirus infections. Unlike standard-of-care nucleoside analogs such as acyclovir or ganciclovir, which require initial phosphorylation by viral kinases (e.g., thymidine kinase), AIC467 directly inhibits the viral DNA polymerase without the need for metabolic activation. This mechanism allows it to maintain potency against resistant viral strains that have lost kinase activity. AIC467 exhibits "pan-herpes" activity, demonstrating effectiveness against multiple members of the Herpesviridae family, including Herpes Simplex Virus 1 and 2 (HSV-1, HSV-2), Varicella-Zoster Virus (VZV), Human Cytomegalovirus (HCMV), Epstein-Barr Virus (EBV), and Human Herpesvirus 6 (HHV-6). It is particularly intended for use in immunocompromised patients who are at high risk for severe or multi-herpesvirus infections.
