AIE-Mit-TPP is a mitochondria-targeting nanomaterial with aggregation-induced emission (AIE) characteristics designed for the treatment of p53-mutated cancers. It selectively targets mitochondria via a triphenylphosphonium (TPP) moiety and functions by eliciting the ubiquitination-dependent proteasomal degradation of a broad range of mutant p53 (mutp53) proteins. This degradation abrogates the neomorphic gain-of-function (GOF) properties of mutp53, which are associated with tumor progression, leading to inhibited cell proliferation, reduced migration, and increased apoptosis. Additionally, AIE-Mit-TPP induces significant mitochondrial damage and triggers autophagy. Research indicates that the inhibition of autophagy can further enhance the mutp53-degrading efficacy of AIE-Mit-TPP, suggesting a potential combination strategy with autophagy inhibitors for optimized cancer therapy. It has demonstrated anti-tumor activity in preclinical models of ovarian cancer.
