Drug intelligence / Profile preview
`AIF-PA1` (Anti-Inflammatory Peptide Amphiphile 1) is a self-assembling peptide amphiphile nanofiber designed for localized anti-inflammatory and regenerative therapy. It consists of a hydrophobic palmitoyl (C16) tail, a beta-sheet-forming peptide segment (VVVAAAEEE), and a bioactive peptide epitope (MQMKKVLDS), which is also known as Antiflammin-1. Antiflammin-1 is a synthetic peptide derived from the region of high sequence similarity between human lipocortin I (annexin A1) and rabbit uteroglobin. Upon self-assembly in aqueous environments, AIF-PA1 forms high-aspect-ratio nanofibers that display the bioactive epitope at high density. AIF-PA1 acts as an inhibitor of phospholipase A2 (PLA2), thereby blocking the synthesis of platelet-activating factor (PAF) and other downstream inflammatory mediators. In preclinical models, local delivery of AIF-PA1 (such as via colonic lavage or scaffold coating) significantly reduces pro-inflammatory cytokines (TNF-alpha, IFN-gamma, IL-1beta), increases anti-inflammatory IL-10, modulates macrophage polarization toward a pro-regenerative M2 phenotype, and promotes tissue repair in conditions like ulcerative colitis, Crohn's disease, urethral defects, and bladder augmentation.
