AIK3a305 is an investigational, orally bioavailable small molecule allosteric inhibitor of p38α mitogen-activated protein kinase (MAPK), developed by Allinky Biopharma. Unlike traditional ATP-competitive inhibitors, AIK3a305 targets an allosteric site on the p38α enzyme, a mechanism designed to provide high selectivity and avoid the off-target toxicities that have historically limited the clinical utility of p38 inhibitors. By modulating the p38 MAPK pathway, the drug aims to suppress the production of pro-inflammatory cytokines such as TNF-α and IL-1β. AIK3a305 is being developed for the treatment of chronic inflammatory diseases, autoinflammatory conditions, and inflammation-related fibrosis. It is currently in Phase 1 clinical development to evaluate its safety, tolerability, and pharmacokinetics in healthy volunteers.
