Overview

A small-molecule agonist of the Absent in Melanoma 2 (AIM2) protein, being developed by IFM Therapeutics for the treatment of colorectal and prostate cancers. AIM2 is a cytosolic DNA-sensing protein that, upon activation, triggers the formation of an inflammasome complex. This leads to the release of pro-inflammatory cytokines such as IL-1β and IL-18 and induces pyroptosis, a form of programmed cell death. In certain cancers, including colorectal and prostate cancer, AIM2 expression is often reduced, suggesting that its activation may be detrimental to tumor survival. The drug is designed to stimulate anti-tumor immunity by recruiting immune cells into the tumor microenvironment and promoting a robust immune response. It is intended for potential use as a standalone therapy or in combination with checkpoint inhibitors to improve treatment efficacy in difficult-to-treat cancers.
