Overview

AIR645 is a second-generation, modified antisense oligonucleotide drug developed for inhalation. It targets the alpha subunit of the human interleukin-4 receptor (IL-4Rα), which is also required for interleukin 13 (IL-13) activity, thereby inhibiting both IL-4 and IL-13 signaling pathways. These cytokines are key mediators in allergic inflammation and asthma pathogenesis. The drug was designed to suppress asthmatic responses by blocking dual cytokine signaling involved in airway inflammation. Developed originally by Isis Pharmaceuticals (now Ionis) and later by Altair Therapeutics, AIR645 reached phase 2 clinical trials for asthma and allergic asthma but was discontinued after these studies[2][3][5].
