Overview

AJMUC1 is an autologous chimeric antigen receptor T cell (CAR-T) therapy engineered to target aberrantly glycosylated mucin 1 (MUC1), a protein overexpressed on the surface of many solid tumors, including breast cancer. The therapy incorporates a knockout of the programmed death-1 (PD-1) gene in the CAR-T cells to prevent immune exhaustion mediated by tumor-expressed PD-L1, thereby enhancing antitumor activity and persistence. The chimeric antigen receptor includes an antigen-binding domain specific for MUC1, a transmembrane domain, co-stimulatory domains, and intracellular signaling domains. AJMUC1 has been evaluated in phase I/II clinical trials for advanced or metastatic breast cancer and was also under development for non-small cell lung cancer and esophageal cancer[4][5][2][6].
