AK007

AK007 is a humanized monoclonal antibody being developed by Allakos Inc. as an antagonist of Siglec-10 (Sialic acid-binding Ig-like lectin 10). Siglec-10 is an inhibitory myeloid checkpoint receptor selectively expressed on tumor-associated macrophages (TAMs) and dendritic cells. By blocking the interaction between Siglec-10 and its known ligands, including CD24, CD52, and VAP-1, AK007 is designed to reverse myeloid-mediated immunosuppression and promote anti-tumor immunity. Preclinical studies have demonstrated that AK007 can polarize tumor-associated myeloid cells toward a pro-inflammatory phenotype and reduce tumor burden, positioning it as a potential therapeutic candidate for various oncology indications.
