Overview

**AK0705** is a potential first-in-class neutrophil elastase inhibitor developed by Ark Biosciences for inflammatory respiratory diseases, primarily **chronic obstructive pulmonary disease (COPD)**. It targets key proteases in the inflammatory pathway of immune cells, forming reversible covalent bonds with exceptional picomolar inhibitory activity. Preclinical studies demonstrated efficacy in alleviating lipopolysaccharide (LPS)-induced acute lung injury (ALI) in mice by reducing proinflammatory cytokines in bronchoalveolar lavage fluid. Phase I clinical trials are planned for 2024 in China, with ongoing co-development by Calibr (Scripps Research).
