Overview

This combination is an investigational therapeutic regimen comprising four agents for oncology use, primarily in advanced or metastatic solid tumors such as gastric or gastroesophageal junction (GEJ) adenocarcinoma. **AK104 (cadonilimab)** is a bispecific monoclonal antibody targeting both PD-1 and CTLA-4, acting as an immune checkpoint inhibitor to enhance antitumor immunity. **AK117 (ligufalimab)** is a humanized IgG4 monoclonal antibody targeting CD47, blocking the CD47-SIRPα interaction to promote macrophage-mediated phagocytosis of tumor cells without causing hemagglutination. **Capecitabine** is an oral prodrug of 5-fluorouracil (5-FU), inhibiting DNA synthesis as an antimetabolite. **Oxaliplatin** is a platinum-based chemotherapeutic causing DNA crosslinking and apoptosis. Clinical trials of this four-drug combination have reported favorable safety and early efficacy signals in patients with advanced HER2-negative gastric and GEJ cancers[2][4]. AK104 and AK117 are both developed by Akeso.
