Overview

This is a **combination regimen** consisting of AK104 (cadonilimab), a bispecific monoclonal antibody targeting both Programmed death-1 (PD-1) and Cytotoxic T-lymphocyte-associated protein 4 (CTLA-4), together with the chemotherapeutic agents oxaliplatin, irinotecan, fluorouracil (5-FU), and leucovorin. AK104 (cadonilimab) acts through immune checkpoint inhibition, potentiating antitumor immune responses. The regimen leverages the immunomodulatory effects of AK104 alongside the cytotoxic action of platinum-based (oxaliplatin) and topoisomerase inhibitor (irinotecan) chemotherapy, as well as the antimetabolite actions of fluorouracil and leucovorin, to treat advanced gastrointestinal and related solid tumors, including colorectal and gastric cancers[3][5][6]. The combination is currently being investigated in clinical trials as neoadjuvant or first-line therapy settings.
