Overview

This combination consists of four anticancer agents: - **AK105** is an anti-PD-1 monoclonal antibody (immune checkpoint inhibitor) designed to block programmed death-1 (PD-1) pathway, enhancing T cell-mediated immune responses against tumors. - **Anlotinib** is an oral multi-targeted tyrosine kinase inhibitor that affects angiogenesis and tumor cell proliferation by inhibiting vascular endothelial growth factor receptors (VEGFR1/2/3), platelet-derived growth factor receptors (PDGFRα/β), fibroblast growth factor receptors (FGFR1-4), c-Kit, and MET[1][3][5]. - **Capecitabine** is an oral prodrug of 5-fluorouracil (5-FU), a cytotoxic antimetabolite interfering with DNA synthesis in rapidly dividing cells[2][6]. - **Oxaliplatin** is a platinum-based chemotherapeutic agent that forms DNA crosslinks, leading to apoptosis of cancer cells[2][6]. The combination brings together immunotherapy (AK105), anti-angiogenic targeted therapy (anlotinib), and cytotoxic chemotherapy (capecitabine, oxaliplatin) for synergistic antitumor effects. It is under clinical investigation for treatment of various advanced malignancies, most notably colorectal cancer and other gastrointestinal tumors.
