Overview

AK112 + AK117 is a combination therapy comprised of Ivonescimab (AK112), a bispecific anti-PD-1/VEGF-A antibody, and Ligufalimab (AK117), a humanized IgG4 monoclonal antibody targeting CD47. AK112 inhibits both PD-1 (a checkpoint pathway controlling T-cell activity) and VEGF-A (a key mediator of angiogenesis), aiming to simultaneously block immune evasion and tumor vascularization. AK117 blocks CD47, a "do not eat me" signal often upregulated in tumor cells, thereby promoting macrophage-mediated phagocytosis of cancer cells. The combination is under clinical development for multiple solid tumors, designed to synergistically activate both innate (via CD47 blockade) and adaptive immunity (via PD-1/VEGF-A inhibition), and is being studied with or without chemotherapy.
