Overview

AK112 + AK117 + chemotherapy is a combination regimen under clinical investigation for advanced solid tumors, especially gastrointestinal cancers (e.g., metastatic colorectal cancer) and possibly other malignancies. AK112 (Ivonescimab) is a bispecific monoclonal antibody targeting both programmed death-1 (PD-1) and vascular endothelial growth factor (VEGF), providing immune checkpoint inhibition and anti-angiogenesis effects. AK117 (Ligufalimab) is a monoclonal antibody that targets cluster of differentiation 47 (CD47), an innate immune checkpoint, to enhance macrophage-mediated phagocytosis of cancer cells. Chemotherapy agents in the regimen frequently include 5-fluorouracil, capecitabine, oxaliplatin, and irinotecan. The combination seeks to activate both innate and adaptive immune responses while inhibiting tumor angiogenesis, potentially providing synergistic antitumor efficacy beyond standard therapies. Both AK112 and AK117 are developed by Akeso. Clinical development is ongoing in multiple cancer types, notably gastrointestinal cancers and head and neck cancers, at the phase Ib/II and phase III trial stage[1][2][3][4].
