Overview

AK112 + etoposide + carboplatin is a combination therapy consisting of ivonescimab (AK112), etoposide, and carboplatin, being investigated primarily for the treatment of advanced non-small cell lung cancer (NSCLC). AK112 (ivonescimab) is a first-in-class bispecific IgG1 monoclonal antibody that simultaneously targets programmed death-1 (PD-1) and vascular endothelial growth factor (VEGF), providing both checkpoint inhibition (immunotherapy) and anti-angiogenic effects. Etoposide and carboplatin are established cytotoxic chemotherapy agents: etoposide inhibits topoisomerase II, causing DNA strand breaks, while carboplatin is a platinum-based agent that forms DNA crosslinks, inhibiting DNA synthesis. The combination aims to enhance antitumor effects by leveraging immunomodulation alongside direct cytotoxicity. The regimen is administered intravenously and is being evaluated in multiple clinical settings, especially in advanced NSCLC both in untreated and pretreated populations[1][5][7].
