AK117 + azacitidine is an investigational combination therapy comprising AK117 (ligufalimab), a next-generation humanized IgG4 anti-CD47 monoclonal antibody, and azacitidine, a hypomethylating agent. AK117 blocks the CD47-SIRPα interaction, thereby promoting phagocytic activity of immune cells against tumor cells. Azacitidine inhibits DNA methylation, interfering with cancer cell growth. This combination is being evaluated in phase 2 clinical trials for the treatment of newly diagnosed higher-risk myelodysplastic syndromes (HR-MDS), aiming to improve remission rates and address unmet needs in this patient population. AK117 is developed by Akeso, which also manufactures the product[1][2][3][7].
