AK117 + nab-paclitaxel + paclitaxel is a combination regimen consisting of three agents: AK117, a monoclonal antibody targeting CD47; nab-paclitaxel (albumin-bound paclitaxel), a nanoparticle formulation of paclitaxel designed to improve solubility and reduce hypersensitivity reactions; and conventional paclitaxel, a widely used antimicrotubule chemotherapeutic. AK117 functions as an immune checkpoint inhibitor by blocking CD47 and enhancing phagocytosis of tumor cells. Nab-paclitaxel and paclitaxel disrupt microtubule dynamics, inhibiting mitosis and inducing apoptosis in cancer cells. This regimen is under investigation for use in solid tumors, based on the rationale of combining targeted immunotherapy (AK117) with two taxanes (nab-paclitaxel and paclitaxel), but is not currently an approved standard regimen.
