Overview

AK119 + AK112 is an investigational combination therapy comprising two monoclonal antibodies developed by Akeso. **AK119** is an anti-CD73 monoclonal antibody targeting the CD73 enzyme, a component of the adenosine pathway that promotes immune suppression within the tumor microenvironment; blockade inhibits adenosine production, potentially restoring antitumor immune responses. **AK112** (also known as ivonescimab or SMT112) is a bispecific monoclonal antibody targeting both programmed death-1 (**PD-1**) and vascular endothelial growth factor (**VEGF**); by simultaneously blocking PD-1-mediated T-cell inhibition and VEGF-driven angiogenesis, it exerts dual immunomodulatory and anti-angiogenic effects. The combination is being developed for advanced solid tumors, including but not limited to microsatellite stable (MSS) / pMMR colorectal cancer and other malignancies, based on studies suggesting synergistic antitumor activity through complementary immune checkpoint blockade and modulation of the tumor environment[1][3][4][6][7][8].
