Overview

AK127 is a humanized IgG1 kappa monoclonal antibody that targets TIGIT (T cell immunoreceptor with Ig and ITIM domains), an immune checkpoint receptor expressed on various immune cells including tumor-infiltrating lymphocytes and natural killer (NK) cells. By binding to TIGIT, AK127 blocks its interaction with ligands such as CD112 and CD155, thereby relieving immune suppression in the tumor microenvironment and promoting anti-tumor immunity. The drug is being developed by Akeso Biopharma for the treatment of advanced solid tumors, including use in combination therapies. Its mechanism of action includes antibody-dependent cellular cytotoxicity as well as stimulation of NK cells and T lymphocytes[1][2][3][4][6][7].
