Overview

AK127 + AK104 is an investigational combination therapy being studied for the treatment of advanced or metastatic solid tumors. Both agents are biologic drugs developed by Akeso. - **AK127** is a monoclonal antibody targeting TIGIT, an immune checkpoint receptor implicated in tumor immune evasion. - **AK104** (also known as cadonilimab) is a humanized tetravalent bispecific IgG1 antibody that simultaneously targets programmed cell death protein 1 (PD-1) and cytotoxic T-lymphocyte antigen 4 (CTLA-4), two key immune checkpoints involved in downregulating anti-tumor immunity[6][8]. The Fc-null design of cadonilimab reduces effector functions such as ADCC and CDC, aiming to minimize toxicity compared to traditional PD-1/CTLA-4 blockade[6]. Preclinical studies have shown that the combination of these two antibodies produces enhanced anti-tumor effects compared to either agent alone, suggesting synergistic activity[5]. The combination is currently being evaluated in early-phase clinical trials for safety, tolerability, pharmacokinetics, immunogenicity, and preliminary efficacy in patients with various advanced solid malignancies[2][3][4].
