Overview

AK131 is a bispecific antibody developed by Akeso that targets both programmed cell death protein 1 (PD-1) and CD73. It is derived from penpulimab (Akeso's marketed PD-1 antibody) and AK119 (Akeso's CD73 antibody in clinical development)[2][3][5]. The drug is designed to block the interaction between PD-1 and its ligands (PD-L1/PD-L2), thereby relieving immune suppression, while also inhibiting the ectonucleotidase activity of CD73 to reduce adenosine accumulation in the tumor microenvironment[5]. This dual mechanism aims to enhance anti-tumor immune responses by overcoming resistance mechanisms associated with single-target immunotherapies. Preclinical studies have shown that AK131 can promote T cell activation and has demonstrated promising anti-tumor activity[2][5]. It is currently being investigated for use in advanced solid tumors, including but not limited to non-small cell lung cancer, esophageal cancer, pancreatic ductal adenocarcinoma, and liver cancer[5].
