AK133 is an investigational bispecific antibody developed by Akeso that simultaneously targets Programmed Cell Death Protein 1 (PD-1) and Human Epidermal Growth Factor Receptor 2 (HER2). By binding to PD-1, AK133 blocks the inhibitory signaling that prevents T-cells from attacking tumor cells, while its HER2-binding arm targets the antibody to HER2-overexpressing cancer cells. This dual-targeting approach is designed to concentrate immune checkpoint inhibition within the tumor microenvironment and promote a more effective, localized anti-tumor immune response. AK133 is being developed for the treatment of HER2-positive advanced solid tumors, aiming to provide a more potent therapeutic option than existing monotherapies or combinations of separate antibodies.
