Overview

AK146D1 is a first-in-class, bispecific antibody-drug conjugate (ADC) developed by Akeso. It is designed to simultaneously target Trop2 (TACSTD2 protein) and Nectin4 (LNIR protein), both of which are highly expressed in various tumors. This bispecific ADC approach aims to improve the specificity and efficacy of treatment by enabling dual targeting within a single molecule, potentially leading to enhanced anti-tumor activity, especially in tumors expressing either or both of these antigens. Preclinical studies have shown strong anti-tumor activity and a favorable safety profile. As of July 2025, AK146D1 is in Phase I clinical trials for advanced solid tumors. It marks the first bispecific ADC targeting both Trop2 and Nectin4 to enter clinical evaluation[1][2][5][7][10].
