Drug intelligence / Profile preview
AK146D1 + Yidafang is an investigational combination regimen consisting of AK146D1, a bispecific antibody-drug conjugate (bsADC) targeting TROP2 and Nectin-4, and Yidafang (ivonescimab), an approved bispecific antibody targeting PD-1 and VEGF-A. Developed by Akeso, this combination is part of the company's 'IO2.0 + ADC2.0' strategy, which aims to combine next-generation immuno-oncology agents with bispecific ADCs to overcome the limitations of conventional therapies. AK146D1 consists of a bispecific antibody targeting TROP2 and Nectin-4 conjugated to a topoisomerase I inhibitor (DXd) payload via a cleavable MC-AAA linker. Ivonescimab simultaneously blocks PD-1 and VEGF-A to restore T-cell activity and normalize the tumor microenvironment. The combination is currently being evaluated in Phase II clinical trials for advanced solid tumors, including non-small cell lung cancer (NSCLC), breast cancer (specifically HR+/HER2- and triple-negative breast cancer), and urothelial carcinoma.
