AK152 is an investigational bispecific antibody developed by Akeso for the treatment of Alzheimer's disease. It is designed with a dual-action mechanism: one arm targets amyloid-beta (Aβ) to facilitate the clearance of neurotoxic soluble Aβ oligomers and insoluble plaques, while the other arm binds to a receptor highly expressed on the blood-brain barrier (BBB). This second arm leverages receptor-mediated transcytosis to significantly enhance the drug's penetration into the central nervous system, addressing a major limitation of conventional monoclonal antibodies. Pre-clinical data suggest that AK152 accelerates Aβ clearance and reverses neuropathology more effectively than standard Aβ-targeting antibodies. It is currently undergoing Phase I clinical evaluation in China to assess safety, pharmacokinetics, and preliminary efficacy in healthy volunteers and patients with mild Alzheimer's disease or mild cognitive impairment.
