AK162 is a first-in-class trispecific antibody developed by Akeso (also known as Akesios). It is designed to simultaneously target three key pathways involved in tumor immune evasion and progression: Programmed Cell Death Protein 1 (PD-1), Vascular Endothelial Growth Factor (VEGF), and T-cell Immunoreceptor with Ig and ITIM domains (TIGIT). By inhibiting the PD-1/PD-L1 axis, AK162 restores T-cell activation; by neutralizing VEGF, it inhibits tumor angiogenesis and reduces immunosuppression in the tumor microenvironment; and by blocking TIGIT, it further enhances the anti-tumor activity of T cells and NK cells. This multi-targeted approach is intended to overcome resistance to current checkpoint inhibitor therapies. AK162 is currently being evaluated in Phase 1/2 clinical trials for the treatment of advanced solid tumors.
