Overview

AK163 is a bispecific monoclonal antibody developed by Akeso (Akeso Biopharma) that targets both Programmed Cell Death Protein 1 (PD-1) and Leukocyte Surface Antigen CD47 (CD47). It is designed to simultaneously inhibit the PD-1/PD-L1 immune checkpoint and the CD47/SIRPα "don't eat me" signaling pathway. By blocking these two pathways, AK163 aims to activate both adaptive immunity (via T cells) and innate immunity (via macrophages) to enhance anti-tumor responses. The dual-targeting approach is also intended to improve the safety profile of CD47 blockade by leveraging PD-1 binding to localize the drug to the tumor microenvironment, potentially reducing off-target effects such as hemagglutination and anemia. AK163 is currently in early-phase clinical development for the treatment of advanced solid tumors and hematological malignancies.
