Overview

AKAP12 shRNA knockdown refers to the experimental use of short hairpin RNA (shRNA) to suppress the expression of the AKAP12 gene, also known as SSeCKS, AKAP250, or gravin. AKAP12 is a hypoxia-inducible scaffold protein that coordinates signaling complexes involving PKC, Src, and F-actin to regulate cell adhesion and motility. While traditionally viewed as a tumor suppressor in various cancers such as prostate and colon cancer, research in melanoma has identified a pro-oncogenic role for AKAP12. Preclinical studies demonstrate that stable knockdown of AKAP12 in melanoma cell lines significantly reduces their invasive and migratory capabilities in vitro and diminishes tumor growth and metastatic potential in orthotopic and metastatic mouse models.
