Overview

*Akkermansia muciniphila* is a species of Gram-negative, anaerobic bacteria that naturally resides in the mucus layer of the human gastrointestinal tract. It is being widely studied and developed as a "next-generation probiotic" or live biotherapeutic product for a range of health conditions. Its abundance in the gut has been inversely correlated with obesity, metabolic syndrome, type 2 diabetes, and certain inflammatory diseases. The bacterium's primary mechanism involves degrading intestinal mucin to produce short-chain fatty acids, such as acetate and propionate, which serve as energy for other gut microbes and the host. This process helps to strengthen the integrity of the gut barrier, reduce low-grade inflammation, and modulate host metabolic and immune responses. Both live and pasteurized (heat-killed) forms of the bacterium have demonstrated beneficial effects, with research suggesting that the pasteurized form may be more potent in some contexts. Specific outer membrane proteins, such as Amuc_1100, are believed to mediate some of its therapeutic effects by interacting with host receptors like TLR2. Several companies, including A-Mansia Biotech (rebranded as The Akkermansia Company), are developing *A. muciniphila* as a nutritional supplement and a pharmaceutical agent for indications including metabolic health, weight management, and as an adjunct to cancer immunotherapy.
