AKP-022 is an investigational, orally active small molecule gonadotropin-releasing hormone (GnRH) receptor antagonist being developed by ASKA Pharmaceutical. It is currently in Phase 1/2 clinical development in Japan for the treatment of hormone-dependent gynecological conditions, specifically uterine fibroids and endometriosis. The drug works by competitively binding to GnRH receptors in the anterior pituitary gland, which inhibits the secretion of gonadotropins, namely luteinizing hormone (LH) and follicle-stimulating hormone (FSH). This suppression leads to a reduction in the production of ovarian steroid hormones, such as estrogen and progesterone, creating a hypoestrogenic environment that inhibits the growth of hormone-dependent tissues like uterine fibroids and ectopic endometrial implants. In clinical trials, AKP-022 is being compared to relugolix, an established GnRH antagonist, to evaluate its efficacy, safety, and pharmacodynamic profile in premenopausal women.
