AKP-201 is an orally bioavailable, selective P2X3 receptor antagonist currently under development by Asahi Kasei Pharma. It is primarily being investigated for the treatment of refractory or unexplained chronic cough (RCC/UCC). The drug's mechanism involves the inhibition of P2X3 receptors, which are ATP-gated ion channels located on sensory nerve fibers (C-fibers) in the airways. In chronic cough conditions, excessive ATP release and subsequent P2X3 receptor activation lead to neuronal hypersensitivity and an exaggerated cough reflex. By antagonizing these receptors, AKP-201 aims to reduce the hypersensitivity of the cough reflex arc and decrease cough frequency.
