AKY-1189 is a novel radiopharmaceutical designed to deliver targeted alpha radiation to tumor cells by binding specifically to Nectin-4, a cell surface protein highly expressed in various solid tumors including bladder (urothelial), breast (including triple negative), lung (non-small cell), cervical, and colorectal cancers. The drug consists of a proprietary miniprotein engineered for high affinity and selectivity for Nectin-4, conjugated with the alpha-emitting radioisotope actinium-225. This approach enables precise delivery of cytotoxic radiation directly into cancer cells while minimizing exposure to healthy tissues. Preclinical and early human imaging studies have demonstrated robust tumor uptake with limited normal tissue accumulation, supporting its potential as an effective therapy for advanced or metastatic solid tumors that express Nectin-4. AKY-1189 is the first clinical candidate from Aktis Oncology’s miniprotein radioconjugate platform[1][2][3][5][6].
