AL-611 is an investigational small molecule prodrug developed as an oral inhibitor of the hepatitis C virus (HCV) NS5B RNA-dependent RNA polymerase. It is a guanosine nucleotide analogue that, upon metabolic activation, potently inhibits HCV replication by targeting the viral NS5B polymerase. In preclinical studies, AL-611 demonstrated high levels of its active nucleoside 5′-triphosphate in primary human hepatocytes and in vivo in dog liver following oral administration. The compound showed potent anti-HCV activity with EC50 values as low as 5 nM in replicon assays. AL-611 was initially developed by Alios BioPharma and later associated with Janssen BioPharma for further research and development. Its clinical development was discontinued due to competitive reasons before reaching advanced clinical phases[1][2][3][4][5][7].
