Al-Cy5.5 is a novel near-infrared fluorescence (NIRF) emitting molecular imaging probe designed to target atherosclerotic calcification. It is synthesized by chemically coupling alendronate, a bisphosphonate with high affinity for calcium phosphate-containing minerals, to the NIRF dye Cy5.5 via an azide-PEG-NHS ester linker. The probe specifically binds to hydroxyapatite and calcified tissues, allowing for the visualization of spotty vascular calcification, which is a marker of plaque vulnerability. In preclinical murine models, Al-Cy5.5 has demonstrated the ability to selectively target and image calcified atheroma in vivo using high-resolution multichannel intravital molecular imaging systems.
