AL002 is a humanized monoclonal IgG1 antibody designed to activate the triggering receptor expressed on myeloid cells 2 (TREM2), an innate immune receptor found on microglia in the central nervous system. By binding to and stimulating TREM2, AL002 aims to enhance microglial activation and phagocytosis of amyloid beta and other damaged proteins, promote microglial proliferation and survival, and regulate inflammatory signaling. The drug was developed for the treatment of early Alzheimer's disease with the goal of slowing disease progression by modulating neuroinflammation and promoting clearance of pathological proteins. Preclinical studies demonstrated that AL002 induced microglial proliferation and reduced amyloid plaques in animal models. In clinical trials, intravenous administration showed target engagement (reduction in soluble TREM2) and evidence of pharmacodynamic activity but did not demonstrate clinical efficacy in phase 2 trials for Alzheimer's disease[3][4][5][6][8][10].
