AL01211 is a potent, selective, and non-brain penetrant small molecule inhibitor of glucosylceramide synthase (GCS), also known as ceramide glucosyltransferase (UGCG). Developed by AceLink Therapeutics, it is designed to treat autosomal dominant polycystic kidney disease (ADPKD) and Fabry disease by reducing the production of glycosphingolipids. Unlike other GCS inhibitors, AL01211 is engineered to distribute readily into peripheral tissues like the kidney while avoiding the blood-brain barrier, thereby minimizing potential central nervous system side effects. It is currently being evaluated in Phase 1 clinical trials for safety, pharmacokinetics, and pharmacodynamics in healthy volunteers and patients with ADPKD, with plans to enter Phase 2 trials for Fabry disease.
