AL137-ABC is a preclinical-stage monoclonal antibody being developed by Alector for the treatment of Alzheimer's disease. It consists of a high-affinity, fully human antibody that selectively targets PyroGlu3, a toxic epitope of amyloid beta (Aβ) found in plaques. The antibody is engineered with Alector's proprietary Alector Brain Carrier (ABC) technology, which utilizes tuned transferrin receptor binding to facilitate transport across the blood-brain barrier, thereby increasing brain penetration while minimizing peripheral hematologic effects. AL137 is designed to promote myeloid-mediated clearance of amyloid plaques through active effector function. Preclinical data suggest robust brain uptake at low doses, potentially allowing for low-dose, subcutaneous administration. The company expects to file an Investigational New Drug (IND) application in late 2026 or early 2027.
