AL28979 is an orally bioavailable small molecule inhibitor that specifically targets the KRAS G12C mutation. Developed by Adcentrx Therapeutics (through its subsidiary Nanjing Aidecheng Pharmaceutical Technology), the compound is designed to treat advanced or metastatic non-small cell lung cancer (NSCLC) and other solid tumors harboring the KRAS G12C mutation. AL28979 works by covalently binding to the cysteine residue at position 12 of the KRAS protein, locking it in an inactive GDP-bound state and thereby inhibiting downstream oncogenic signaling pathways such as MAPK/ERK. This inhibition leads to the suppression of tumor cell growth and survival. The drug is currently being evaluated in a Phase I clinical trial to determine its safety, tolerability, pharmacokinetics, and preliminary antitumor activity in patients with KRAS G12C-mutated cancers.
